very good question. in fact i have taken this information from a book on pronunciation entitled: "Phonology in English ******** Teaching" by M. C. Pennington.
the best way to pronounce this long word should be done in the following way

pneumonoultramicroscopicsilicovolcanoconiosis

the strong syllables are written in big letters. try to pronounce them louder with higher pitch. the last underlined syllable must receive the strongest stress.
